Can a computer that supports PCI Express x16 use a PCI Express 2.0 X16 video card?

what is the difference between the two?|||No. If you had a PCIE card and your board was PCIE 2 or 3 then it would work. You can't use a technology that comes later in one that came prior. IE: You can use 1.0 on a 2.0 mobo but you cannot use a 2.0 on a 1.0 mobo.|||PCI Express 2.0 means you have support for multi-gpu setups BOTH running at 16x, as opposed to one running at 16x, and the second running at 8x.

This question only applies if you're using a GX2 series or X2 series cards, which are really crossfired or SLI boards on a single card. In THAT case, it would apply and be a problem, maybe. Chances are, the 2.0 version is just an advertising point to say that it's newer. It should work.
